So, going behind my 427 is a custom built 700R4. I need to know exactly how far back I need to move the tranny crossmember. I have heard 3 inches. Is this correct? Please chime in. Need it for Sunday.

I think your best bet would be to bolt it up to the motor, have the crossmember bolted on to the trans, and lift it up to the frame with a jack and then drill the holes in the frame. I wouldn't drill any holes in advance expecting them to line up. Just get it bolted up and see where the holes need to be and you should be good to go.

I just did mine (not a big block) but a T350 to 700r4 swap. Mine is a 4x4 and I used the spacer type adapter. I was able to put the front cross member bolt in the rear cross member bolt hole and drill only one new hole per side.
